Wow, I didn't think you'd actually respond! It is disappointing that the base game might have a smaller cast than what the first game had at launch, but I do understand why, after looking into what you are planning. There's a bunch of talented people around that grew up with that first game, so I'm sure you'll find people capable of helping you.

(before you ask me, I'd love to help, but I gather you're looking for programmers, and I am a lousy programmer. I can do business management and I can do story writing, but I don't think that's quite what you're looking for at the moment)

As much as it hurts to see cut characters its probably for the best. I would love to see them return as DLC but for the sake of getting the game to casual players its for the best.

I think the main returning cast would be Pico, Tankman, P-Bot, Hank, Alien Hominid, and Fancy Pants. You could cut Salad Fingers for DLC since Salad Fingers is important to Newgrounds and is popular since people would be willing to pay money for popular characters.

However I feel like your inclusion of characters you've added are all from games so to balance it out why not add characters from animated series or old Newgrounds mascots. Bitey, Ena, Shotgun Man, Edd, Blockhead, Master Mind, Level 58, and Larry. This isn't limited to flash games, its a celebration of Newgrounds as a whole.

It hurts to actually bump this topic, but I gotta give it my thoughts. Personally a NGR2 would be cool. I do hope that it has the traditional fighting game style (eg Street Fighter, Guilty Gear).

And yeah, I do expect Boyfriend if we're ever gonna see one. But I REALLY want to also see Xiao Xiao, Sonny, and the protagonist from My Friend Pedro. Maybe give them something for old and new fans alike.
